From 2pm councillors at Eastleigh Borough Council (EBC) will discuss the proposals to expand the runway by 164m.
The full council meeting will be held online - links to watch different sections of the meeting using a web browser are available from this EBC web page.
Definite timings are yet to be provided.
But the borough council said it is "likely" that the final decision will either be made very late on Thursday evening or Friday morning, or the council meeting will reconvene on Friday at 6pm.
As previously reported, the plans were rejected by EBC's Eastleigh Local Area Committee on March 25 after a meeting that lasted almost 17 hours, on the grounds of climate change, noise and pollution.
An announcement was then read out by the local committee's chair – who subsequently resigned from the ruling Lib Dem group - that the final decision had to go to EBC's full council to determine due to the application having a "significant borough wide impact".
According to the borough council, more than 200 people have registered to make representation at tomorrow's meeting.
The head of housing and development at Eastleigh Borough Council is expected to present the plans at the start of the meeting and answer initial clarification from councillors.
Members will then hear comments from those against the proposals, from those in favour and from Southampton Airport bosses.
Councillors will then have the opportunity to ask further questions and clarification to the planning officer before a debate begins.
According to the council, the debate will be limited to five minutes per councillor.
At the end of the debate, all 38 councillors will be asked to vote.
Their decision could result in the plans being approved, rejected or deferred.
Both an approval or rejection could be challenged, meaning the plans would then be considered by a planning inspector.
If the decision is deferred, it will be considered by full council in Eastleigh at a later date.
As previously reported, planning officers have recommended for the plans to be approved.
